Lectins are carbohydrate-specific proteins that act as recognition molecules in various biological systems. These proteins bind reversibly to carbohydrates and are highly specific for monosaccharides/oligosaccharides and are also known as glycan-binding proteins. CD BioGlyco utilizes Cu(I)-assisted 1,3-dipolar azide-alkyne cycloaddition (CuAAC) reactions to provide development services for different reagents through different strategies.
Here, we utilize click chemistry methodology to provide a trusted carbohydrate click-based lectin ligand development service.
- We use aromatic compounds and aldehydes as the main sources for the two-step preparation of stereoselective aromatic compounds. We characterize them using nuclear magnetic resonance techniques and infrared spectroscopy.
- Next, we couple aryl hydroxylated glycosides to azide-functionalized glycosides via Cu(I)-assisted 1,3-dipolar azide-alkyne cycloaddition (CuAAC) to obtain a high yield of aryl hydroxylated glycoclusters, which are used as ligands for subsequent binding to the lectin receptor. In this process, the CuAAC reaction is characterized by high efficiency and yield and avoids incomplete reaction of alkyne groups.
- Then, we bind the prepared glycoclusters to the carbohydrate epitopes of lectins to research the binding effect of glycoclusters on lectins. Among them, we provide hemagglutination inhibition assay (HIA), enzyme-linked lectin assay (ELLA), and surface plasmon resonance technique (SPR) to research the binding affinity of glycocluster to lectin.
- HIA: We separately co-incubated different amounts of lectin solution with diluted glycoclusters. After some time, erythrocyte solution is added to it and the incubation is continued. Finally, the lowest inhibitory concentration of each glycocluster molecule is determined.
- ELLA: We use carbonate buffer solution to dissolve the glycocluster ligand and add biotinylated lectin solution to it for co-incubation. After some time, the peroxidase coupling material is added and the absorbance is measured to determine the semi-inhibitory concentration of the glycocluster ligand.
- SPR: A biotinylated glycan solution is applied to the surface of the lectin by streptavidin-biotin interaction, followed by inhibition experiments.
- Furthermore, we provide molecular modeling to research the binding sites of glycocluster and lectin. Combined with the use of fluorescent labeling, the exact location of the glycocluster as it binds to the surface of the lectin is determined in a model of the lectin's steric structure.

Results: In this article, the authors describe the synthesis of a family of tetravalent galactose- and lactose-functionalized macrocycles based on the resorcin aromatic core. Next, the authors also describe the diastereoselective synthetic pathway for cyclopropargylated resorcinol alkenes and their functionalization via copper-catalyzed azide-alkyne click chemistry. Research on ELLA binding confirmed that galactose glycosyl clusters are potent ligands for the bacterial lectin of Pseudomonas aeruginosa, and bind poorly to lactose-based monovalent probes, which also bind poorly to polyvalent glycoclusters of the human galactose lectin.
