Results: This article discusses specific and quantitative methods for biomolecular labeling using click chemistry. It is pointed out that the most commonly used quantitative bioanalysis method is the expression of foreign proteins using fused fluorescent tags. There are two challenges: the stoichiometry of precisely controlling chemical reactions in living cells and the volume of fluorescent labels limit the analysis of the inherent physical and biological properties of proteins. Click chemistry specifically and stoichiometrics label endogenous proteins or other biomolecules within cells. These chemical reactions take advantage of biological orthogonality, mild reaction conditions, high reaction ratio, small functional group size, stable covalent bonds, and simple metabolic labeling using various biomolecular analogs in living cells. These methods have been used in various studies in the field of quantitative biology, such as quantification of newly synthesized proteins and observation of post-translational modifications. Through click chemistry, quantitative fluorescence labeling of biomolecules can be realized for observing biological phenomena and studying systems biology.
