Results: This article mainly discusses a combination of two chemical methods for the labeling of glycans in living cells. The researchers used the Diels-Alder reaction and click chemistry (SPAAC) to simultaneously detect two different metabolic sugars. By adding Ac4ManNPtl, which contains an olefin label, and N-acetyl compounds, which contain an N-azidoacetyl label, to HeLa S3 cells, they successfully labeled two different sugars on the cell surface. The researchers also found that the olefin label had less background staining in the cells and that the non-specific staining could be due to non-specific binding or the addition reaction of the SH group to the triple bond. In addition, the researchers found that this method can be used simultaneously with sugars containing alkyne labels, enabling simultaneous detection of two different sugars. This method is important for advanced multicarbohydrate analysis. In addition, because olefin labeling can also be easily accepted by other biomolecules (such as proteins), this labeling method is not only suitable for metabolic glycan engineering but also can be widely used in other fields.
