  • First, we synthesize unnatural amino acids with alkyne groups by chemical synthesis. These unnatural amino acids contain alkyne groups, azide groups, and other functional groups that are necessary to click reactions and can be selectively captured by another compound containing electrophilic reactive functional groups.
  • Then, we introduce genes for unnatural amino acids containing alkynyl groups into host cells to express the target proteins in the cells. The target proteins are collected from the cells and undergo a series of purification steps to obtain highly purified target proteins.

The modified proteins are then analyzed using mass spectrometry and electrophoresis to determine the location and extent of modification.

Results: This article focuses on the advances in chemical protein modification. Specifically, the article explores chemical methods and techniques for protein modification and the application of these methods in biomedical research and drug development.
