Click Chemistry Technology is a method of linking molecules through covalent bonds and is used to synthesize glycosides. Glycosidation changes the biochemical properties of the substrate and improves its solubility, stability, or bioavailability. Therefore, glycosides have a wide range of applications, including medicine, food, agriculture, etc. We mainly carry out efficient synthesis of glycosides through the following steps.
We select compounds with acceptable functional groups (such as alcohols, amines, etc.) as substrates and sugar molecules as sugar donors.
We consider the compatibility of functional groups when designing click reactions and introduce appropriate clickable functional groups (such as alkynyl groups, azide groups, or similar functional groups) into the substrate and sugar donor to facilitate click reactions.
We mix a substrate with clickable functional groups and a sugar donor together and add a suitable catalyst to promote the formation of a stable glycosidic bond at an appropriate temperature and reaction time to synthesize the target glycoside.
Click chemistry technology has high selectivity, high efficiency, and wide applicability in glycoside synthesis. We provide custom synthesis for various types of glycosides by selecting appropriate click reactions according to different substrates and sugar donors. Results: The authors developed a mild and efficient stereoselective method for the synthesis of C-alkynyl glycosides. The authors used TMS-protected C-alkynyl-glycosides and organic compounds R-N3 as raw materials and performed Cu(I)-promoted azide-alkyne 1,3-dipole ring click reaction to synthesize functionalized C-glycosyl 1,2,3-triazole, and the reaction process was accelerated by ultrasonic irradiation.
