Click Chemistry Technology is a method of linking molecules through covalent bonds and is used to synthesize glycosides. We consider the compatibility of functional groups when designing click reactions and introduce appropriate clickable functional groups (such as alkynyl groups, azide groups, or similar functional groups) into the substrate and sugar donor to facilitate click reactions.

  • Click reaction

We mix a substrate with clickable functional groups and a sugar donor together and add a suitable catalyst to promote the formation of a stable glycosidic bond at an appropriate temperature and reaction time to synthesize the target glycoside.

Results: The authors developed a mild and efficient stereoselective method for the synthesis of C-alkynyl glycosides. The authors used TMS-protected C-alkynyl-glycosides and organic compounds R-N3 as raw materials and performed Cu(I)-promoted azide-alkyne 1,3-dipole ring click reaction to synthesize functionalized C-glycosyl 1,2,3-triazole, and the reaction process was accelerated by ultrasonic irradiation.
