Properties
Description
Desthiobiotin is a sulfur-free analog of biotin, featuring a single-ring structure. It exhibits nearly equal specificity but lower affinity for streptavidin compared to biotin. As a result, desthiobiotinylated proteins can be efficiently eluted from streptavidin affinity resin under mild conditions via- competitive displacement with free biotin.
Molecular Formula
C23H38N4O9
Functional Group 1
Desthiobiotin
Solubility
Water, DMSO, DCM, DMF
Identity
Confirmed by NMR.
Applications
Desthiobiotin is widely used in pull-down experiments with biological samples, offering a unique soft-release property. This characteristic enables the specific elution of desthiobiotinylated proteins from streptavidin resin using free biotin. The gentle elution conditions minimize the co-purification of endogenous biotinylated molecules, which remain bound to streptavidin. This feature is particularly advantageous for preserving the integrity of protein complexes and reducing background noise in downstream applications such as mass spectrometry analysis and protein-protein interaction studies.
